Ms. Miyamoto has really challenged the definition that many have of what it means to be Japanese.
この文章のofの役割が、よくわかりません。
「何をもってして日本人であることを意味するのかという、多く人が抱く定義に対して、宮本さんは挑戦した」というような意味であっていますか？

Ms. Miyamoto has really challenged the definition that many have of what it means to be Japanese.

the definition が先行詞で目的格の関係代名詞の that を用いて many (people) have ～が修飾していく
ということはおわかりだと思います。

ここでは先行詞は本来、the definition of what it means to be Japanese という長いもので、
that I have のみで修飾、と言ってもいいのですが、英語は後ろ後ろへと延びていく言語なので、
the definition / of ～と分けて、the definition that I have / of ～としています。

すなわち、the definition of what it means to be Japanese を考えればいいです。
the definition of ～で「～の定義」

it は形式主語で＝to be Japanese「日本人であること」
what は「もの、こと」という関係代名詞、あるいは「何を」という疑問詞どちらでもいいです。

「日本人であることが意味することの定義」
「日本人であることが何を意味するかの定義」

定義と mean が redundant な気もします。

of は「～という」と同格的にとらえてもいいですので
おっしゃるような訳でいいと思います。
ただ、challenge は「疑問に思う、意義を唱える」くらいがいいかと思います。

　解釈はそれでいいと思います。(「宮本さんは、日本人はこういうものだという通念に一石を投じたのです」 なんて訳し方もあるかもしれませんが ・・・)

　of については、the definition (that many have) of what it means to be Japanese としてみると分りやすくないでしょうか。

　→ the definition of what it means to be Japanese

　こういう文として読んで、それから ( ) の中に入れておいた部分を入れて改めて読んでみると、つながりが分りやすいかと思います。

　つまり、その of は、the definition of ... というつながりにおける of なのです。
