You are an old friend of the bridegroom's.（君は新郎の旧友だからね。）という例文で、なぜbridegroom'sとなっているのか分かりません。'sは必ず必要なのですか？bridegroomではダメですか？

I-my-me-mine, you-your-you-yours 　 etc.
は、それぞれ代名詞の、「主格」「所有格」「目的格」「独立所有格」いいます。「独立所有格」は「～のもの」という意味を持っています。
my, your, his, her, their, its という代名詞の所有格と、a, the の冠詞、this, that などの指示代名詞、some, many などの形容詞とは現代英語では一緒に用いないという約束があります。昔は用いていたこともあるようです。例えば、

He is a my friend.（×）
I know this her friend.（×）
I saw some your friends.（×）

なのです。これは正しく書けば、

He is a friend of mine.
I know this friend of hers.
I saw some friends of yours.
(I saw some of your friends.)
となります。mine, hers, yours は上で述べましたように、独立所有格というものです。

mine は、「私のもの」＝「私の友達」＝my friends
hers は、「彼女のもの」＝「彼女の友達」＝her friends
yoursは、「あなたのもの」＝「あなたの友達」＝your friends
を表しています＃１の方が述べられていますように、of mine, of hers, of yours の形がポイントです。

aとmy（所有格）が同じ名詞を修飾するとき
以下のような文はまちがいです。

He is a my new freind.
彼は新しい私の友だちです。

どう見ても見慣れませんよね。
上記は間違いです。

では、どうなると正しいかというと、

He is a new friend of mine.

となります。
of mineがポイントです。

というわけで、bridegroom'sとなるワケが
推察できたでしょうか？

mine 同様、bridegroom'sは所有格なのです。
bridegroom'sは、「新郎の」、ではなく、
「新郎のもの」という所有格なのです。

＞You are an old friend of the bridegroom's

この文章はanとtheが入り混じっているためにこういう言い回ししているんでしょうね。

You are an old bridegroom's friend.

というのも合っていると思います（→確かめていないので確実ではありません）
